Neurenzin-2 je protein koji je kod ljudi kodiran genom NRSN2.[5][6]
Kloniranje i ekspresija
Koristeći BLAST analizu sa sekvencom mišjeg neurezina-1 (NRSN1; 616630) kao upit, Nakanishi et al. (2006) su identifikovali sekvence mišjeg i ljudskog homolognog gena koje su nazvali neurenzin-2 (NRSN2). Ljudski protein NRSN2 sadrži dva domena koja se protežu kroz membranu i dijeli 41% identiteta s mišjimm Nrsn1. Northern blot analiza mišjih tkiva pokazala je specifičnu ekspresiju za mozak 1,7-kb Nrsn2 transkripta. Imunoblot analiza pokazala je ekspresiju proteina Nrsn2 15,5 embrionskog dana, sa snažnom ekspresijom viđenom od 7 -14 postnatusnih dana i u dobi od 10 sedmica. Imunoblot analiza moždanog područja pokazala je da je Nrsn2 široko eksprimiran kroz mozak, sa najvećom ekspresijom u talamusu/hipotalamusu. Indirektna imunofluorescencija u transficiranim Neuro2a ćelijama pokazala je da se Nrsn2 lokalizira u perijedarne organele , sa procesima u regijama različitim od onih obojenih Nrsn1, a u transformiranim COS-7 ćelijama pokazalo je da se Nrsn2 lokalizira u lizosomima. Imunohistohemijska analiza u mozgu miša pokazala je sličnu ukupnu raspodjelu Nrsn2 i Nrsn1, ali različite subćelijske lokalizacije, s Nrsn2 u somatskim regijama i Nrsn1 u neuritskim procesima. Nrsn2-pozitivne ćelije su viđene septumskom jezgru, dijagonalnom pojasu, hipokampusu, amigdaloidnom jezgru, jezgru habenula i pokrovi moždane kore, a istaknuto imunobojanje viđeno je u ćelijskim tijelima cerebelarnih Purkinjeovih ćelija.[7]
