Die Gemeinde Menestreau liegt im Nivernais, etwa 46 Kilometer nordnordöstlich von Nevers. Nachbargemeinden von Menestreau sind Entrains-sur-Nohain im Norden und Nordosten, La Chapelle-Saint-André im Osten und Südosten, Menou im Süden, Couloutre im Westen sowie Ciez im Nordwesten.
