Toloksaton adalah antidepresan yang diluncurkan pada tahun 1984 di Prancis oleh Sanofi Aventis untuk pengobatan depresi. Obat ini dihentikan produksinya pada tahun 2002.[1] Obat ini bertindak sebagai penghambat MAO-A (RIMA) reversibel selektif.[2]

Sintesis

Reaksi antara glisidol (1) dan m-toluidina (2) menghasilkan 3-m-toluidina-propana-1,2-diol (3). Perlakuan dengan dietil karbonat (4) dengan adanya natrium metoksida menyebabkan sikloadisi intermolekul menghasilkan tomoksaton.[3][4][5]
